I would do it with a dremel small hole first from the inside. Then folowing it with a drill press on the out side of the mold. Use metal for the dowels(A bolt cut down in a drill like a lath with a grinder.) The nice thing is if it is off a little you still have your own design and it might work different. The other way would be what ledhed recommends. But you can ruin a lot of molds for the same cost. Good luck I love to modify or cut my own.
